Androgen Receptor Partial Agonism
RAD-140 binds the androgen receptor with a Ki of approximately 7 nM, acting as a potent partial agonist in anabolic tissue and a partial antagonist in reproductive tissue. This tissue-selective AR modulation is mediated by differential co-activator and co-repressor recruitment upon ligand-induced receptor conformational change.
PI3K/Akt/mTOR Pathway Activation
AR agonism by RAD-140 in skeletal muscle promotes downstream activation of the PI3K → Akt → mTOR signaling axis. This pathway drives protein synthesis, suppresses autophagy, and promotes myofibrillar anabolism — effects studied extensively in models of muscle wasting and age-related sarcopenia without the androgenic side-effect profile of steroidal compounds.
Neuroprotective AR-Mediated Signaling
