What Is Selank?
Selank is a synthetic heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. It was rationally designed as a metabolically stable analog of tuftsin — a naturally occurring immunoregulatory tetrapeptide (Thr-Lys-Pro-Arg) found in the heavy chain of human immunoglobulin G (IgG). Selank was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences in collaboration with the V.V. Zakusov Research Institute of Pharmacology, Moscow.
The C-terminal Pro-Gly-Pro (PGP) tripeptide extension — the same structural motif found in Semax — serves two purposes in preclinical research: it enhances the peptide's metabolic stability against enzymatic degradation, and it is hypothesised to facilitate central nervous system delivery via lipid raft-mediated and receptor-mediated transcytosis mechanisms at the blood–brain barrier. Without this extension, the parent tuftsin tetrapeptide is rapidly degraded by serum carboxypeptidases.
Selank has completed Phase III clinical trials in Russia for generalised anxiety disorder and neurasthenia, where it is approved as a 0.15% intranasal formulation under the name Selank. As of April 2026, it is not FDA-approved for any use in the United States and is sold by PureRawz exclusively as an in-vitro research reagent. All preclinical findings cited on this page arise from animal or cell-based models.
In published literature, Selank is studied across five primary research domains: GABAergic neurotransmission, BDNF expression and neurotrophin modulation, enkephalin-degrading enzyme inhibition, serotonergic and dopaminergic pathway interactions, and immune system gene expression. Its pharmacological distinction from classical benzodiazepines — anxiolytic activity without sedation, tolerance, or dependence in preclinical models — makes it a useful tool for dissecting these pathways independently.
Mechanism of Action — Six Research Pathways
Peer-reviewed preclinical literature has characterised at least six distinct molecular pathways. All findings are from in-vitro or animal studies only.
Selank allosterically modulates GABA-A receptors, increasing inhibitory GABA neurotransmission. Real-time PCR analysis of 84 neurotransmission genes in rat frontal cortex found significant expression changes in 45 genes at 1 h post-administration, with a strong positive correlation between Selank and exogenous GABA effects — positioning Selank as a non-classical GABA-A receptor modulator.

Selank has been shown to upregulate BDNF mRNA in the rat hippocampus and prefrontal cortex following intranasal administration. In models of ethanol-induced memory impairment, Selank's cognitive-protective effects correlated directly with BDNF content normalisation in these regions — connecting the neurotrophin axis to the peptide's nootropic profile.

Selank inhibits enkephalinase — the serine protease responsible for breaking down enkephalins in serum and CNS tissue.
